We develop a new deconvolution approach, where we allow for a high level of dynamics in signature matrix S, except for retaining its structural composition learnt from large amount of training data sets as well as the tissue expression data itself. The unbiasedly estimating proportion of immune and stromal cells could then be reliably correlated with features derived from imaging, genomic and clinical data of these patients.
